OSRM computes shortest or fastest routes on an OpenStreetMap road network, in milliseconds even across a whole country. Beyond a single route it produces distance and time matrices between hundreds of points, solves vehicle routing problems, and computes service areas (isochrones).

In engineering that lets you compute ambulance or fire engine response times from every point in a city, plan waste collection or delivery rounds, and build origin-destination matrices for transport analysis.

What is missing: like OpenTripPlanner, it is a service you install and run yourself, not a desktop application.
